Tebenafusp represents an important advance in the treatment of metastatic uveal melanoma and is the first systemic therapy to demonstrate a significant overall survival benefit in this disease. Its novel mechanism of action offers a new therapeutic approach for selected patients with metastatic uveal melanoma.
Tebentafusp is an innovative immunotherapy that redirects T cells towards uveal melanoma cells by targeting the melanoma-associated protein gp100. It is indicated for patients with metastatic uveal melanoma who are HLA-A*02:01 positive.
In the phase 3 IMCgp100-202 trial, tebentafusp significantly improved overall survival compared with the control group. Median overall survival was 21.7 months with tebentafusp compared with 16.0 months in the control group, with the survival benefit maintained during longer-term follow-up.
This is particularly important because effective systemic treatment options for metastatic uveal melanoma have historically been limited. Tebentafusp has therefore become an important component of the modern management of eligible patients with metastatic disease.
